Watanabe Poemi ist ein fast normales Mädchen. Sie wünscht sich nichts sehnlicher als Seiyuu zu werden und würde für diesen Job alles tun. Leider muss sie sich erst mal um die Rettung der Erde kümmern, denn Aliens haben große Pläne und wollen die Herrschaft an sich zu reißen. Glücklicherweise gibt es die Aasu-Girls, Schwestern, die sich geschworen haben ihre Kräfte für das Wohl der Menschheit einzusetzen. Zu dumm, dass diese Kräfte wie übermenschliche Geschwindigkeit oder das Herbeizaubern von Blumen völlig nutzlos sind. Eine der Aasu-Schwestern ist Poemis Mitschülerin Futaba, die sie bei sich aufnimmt, als auch Poemis Eltern den Aliens zum Opfer fallen. So kommt es, dass Poemi ihren Teil tun will und sich mit Hilfe eines Fisches in das Magical Girl Puni Puni Poemi verwandelt.
